چکیده

The objective of the study was to determine the effect of combined treatment with IEM-1460 (a selective GluR2lacking, Ca2+ permeable AMPA receptors) and conventional antiepileptic drugs: CBZ (carbamazepine), valproate (VPA), phenobarbital (PB), and phenytoin (PHT) against maximal electroshock-induced convulsions in mice. IEM1460 (up to 30 mg/kg) did not show anticonvulsant action in the test for electroconvulsive threshold. The substance did not influence the antiseizure effectiveness of antiepileptic drugs. This may indicate that not all AMPA receptor antagonists may be useful as additive drugs in the complex therapy of epilepsy. On the other hand, the GluR2-lacking, Ca2+ permeable subtype of AMPA receptors may be not crucial for seizure phenomena.

The role of BK channels in antiseizure action of the CB1 receptor agonist ACEA in maximal electroshock and pentylenetetrazole models of seizure in mice

The anticonvulsant effect of cannabinoid compound has been shown in various models of seizure. On the other hand, there are controversial findings about the role of large conductance calcium-activated potassium (BK) channels in the pathogenesis of epilepsy. In this study, the effect of arachidonyl-2′-chloroethylamide (ACEA), a CB1 receptor agonist, and a BK channel antagonist, paxilline, either...

Evaluation of GluR2 subunit involvement in AMPA receptor function of neonatal rat hypoglossal motoneurons.

AMPA receptors (AMPAr) mediate fast synaptic responses to glutamate and, when they lack the GluR2 subunit, are strongly Ca2+ permeable and may increase intracellular Ca2+ levels. Because hypoglossal motoneurons possess restricted ability to buffer internal Ca2+ and are vulnerable to Ca2+ excitotoxicity, we wondered if, in these cells, any significant Ca2+ influx could be generated via AMPAr act...

Effect of MPEP, a selective mGluR5 antagonist, on the antielectroshock activity of conventional antiepileptic drugs.

MPEP, a selective non-competitive antagonist of group I metabotropic glutamate receptor subtype 5 (mGluR5), administered at doses ranging from 0.75 to 1 mg/kg, failed to influence the electroconvulsive threshold in mice. However, when administered at higher doses (1.25 and 1.5 mg/kg), it significantly increased the threshold. Moreover, MPEP (applied at its highest subprotective dose of 1 mg/kg)...

Indapamide enhances the protective action of carbamazepine, phenobarbital, and valproate against maximal electroshock-induced seizures in mice.

PURPOSE To determine the influence of indapamide on the protective action of numerous conventional and second-generation antiepileptic drugs (carbamazepine, lamotrigine, oxcarbazepine, phenobarbital, topiramate and valproate) in the mouse maximal electroshock seizure model. MATERIAL AND METHODS Electroconvulsions were evoked in Albino Swiss mice by a current (sine-wave, 0.2 s stimulus duratio...
